Obiettivi

L’insegnamento si propone di sviluppare nello studente la comprensione dei meccanismi di base che regolano l’organizzazione molecolare, le reazioni biochimiche ed i cicli metabolici correlati dai loro principali sistemi di regolazione.

Contenuti sintetici

Lo studente apprenderà 1) le generalità sulle molecole componenti la materia vivente; 2) la struttura, la funzione, il meccanismo d'azione degli enzimi ed il loro ruolo nella regolazione metabolica; 3) il meccanismo con cui l’organismo vivente produce energia; 4) gli aspetti nutrizionali come fonte d’energia nella vita di tutti i giorni e nell’ esercizio fisico; 5) i processi digestivi, le molecole implicate nel metabolismo energetico.

Programma esteso

Introduzione al corso e generalità sulla materia vivente: Introduzione alla Biochimica e alle caratteristiche fondamentali della materia vivente. Organizzazione delle biomolecole e principi generali delle trasformazioni biochimiche.
Biochimica strutturale: Struttura e proprietà delle principali biomolecole: glucidi, lipidi, proteine e nucleotidi.
Enzimologia: Reazioni biochimiche, enzimi, cinetica enzimatica (Vmax e Km) e principali meccanismi di regolazione dell’attività enzimatica.
Bioenergetica: Principi generali della bioenergetica cellulare. Catena respiratoria mitocondriale e fosforilazione ossidativa.
Metabolismo energetico: Principi generali del metabolismo energetico. Valore calorico dei nutrienti, equivalenti calorici e utilizzo dei diversi substrati energetici. Digestione e assorbimento: Principi generali della digestione e dell’assorbimento dei nutrienti.
Metabolismo dei carboidrati: Digestione, assorbimento e trasporto del glucosio e degli altri monosaccaridi. Glicolisi: tappe fondamentali, bilancio energetico e regolazione. Destino del piruvato in condizioni aerobiche e anaerobiche (fermentazione lattica). Glicogenosintesi e glicogenolisi e loro regolazione ormonale. Gluconeogenesi: principi generali e ruolo nel mantenimento della glicemia. Via dei pentoso fosfati: funzioni principali (produzione di NADPH e ribosio-5-fosfato). Integrazione del metabolismo glucidico nello stato alimentato e di digiuno.

Aims

The course aims to teach the basic mechanisms that regulate the molecular organization, the biochemical reactions, the cellular and sub-cellular morphology and the metabolic pathways.

Contents

The student will learn 1) the general information on the molecules that make up living matter; 2) the structure, function, mechanism of action of enzymes and their role in metabolic regulation; 3) the mechanism by which the living organism produces energy; 4) nutritional aspects as a source of energy in everyday life and in physical exercise; 5) digestive processes, the molecules involved in energy metabolism.

Detailed program

Course introduction and general concepts of living matter: Introduction to Biochemistry and to the fundamental characteristics of living matter. Organization of biomolecules and general principles of biochemical transformations.
Structural Biochemistry: Structure and properties of the main biomolecules: carbohydrates, lipids, proteins, and nucleotides.
Enzymology: Biochemical reactions, enzymes, enzyme kinetics (Vmax and Km), and main mechanisms of regulation of enzymatic activity.
Bioenergetics: General principles of cellular bioenergetics. Mitochondrial respiratory chain and oxidative phosphorylation.
Energy metabolism: General principles of energy metabolism. Caloric value of nutrients, caloric equivalents, and utilization of different energy substrates.
Digestion and absorption: General principles of nutrient digestion and absorption.
Carbohydrate metabolism: Digestion, absorption, and transport of glucose and other monosaccharides. Glycolysis: key steps, energy yield, and regulation. Fate of pyruvate under aerobic and anaerobic conditions (lactic fermentation). Glycogen synthesis (glycogenesis) and glycogen breakdown (glycogenolysis) and their hormonal regulation. Gluconeogenesis: general principles and role in maintaining blood glucose levels. Pentose phosphate pathway: main functions (production of NADPH and ribose-5-phosphate). Integration of carbohydrate metabolism in the fed and fasting states.
Overview of lipid and protein metabolism: General principles of lipid and protein metabolism.
