- Introduzione
Syllabus del corso
Obiettivi
Il modulo di chimica biologica fornisce competenze sulle metodologie biochimiche di base, riguardanti tecniche di purificazione, caratterizzazione qualitativa e quantitativa di proteine e attività enzimatiche.
- Conoscenza e capacità di comprensione
Al termine dell’insegnamento lo studente si sarà appropriato delle conoscenze fondamentali per la purificazione e caratterizzazione funzionale di un enzima a partire da un pellet di cellule.
- Capacità di applicare conoscenza e comprensione
Al termine dell'insegnamento lo studente dovrà essere in grado di applicare le conoscenze acquisite nel campo della biochimica.
- Autonomia di giudizio
Lo studente dovrà essere in grado di elaborare quanto appreso e saper riproporre le metodiche utilizzate e quando queste devono essere applicate.
- Abilità comunicative
Alla fine dell’insegnamento lo studente saprà esprimersi in modo appropriato nella descrizione delle tecniche imparate con proprietà di linguaggio e sicurezza di esposizione.

Alla fine dell’insegnamento lo studente sarà in grado di riproporre le tecnologie apprese in contesti analoghi.
Contenuti sintetici
Purificazione e caratterizzazione di una proteina: cromatografia di affinità, dosaggio della concentrazione proteica, valutazione della purezza e determinazione della massa molecolare della proteina purificata. Presentazione di metodi per lo studio della cinetica enzimatica.
Programma esteso
l modulo di chimica biologica prevede 5 esercitazioni pratiche in laboratorio:
1. Preparazione dell’estratto grezzo della proteina di interesse;
2. Purificazione dell’enzima tramite cromatografia di affinità e dosaggio della concentrazione proteica mediante saggio colorimetrico;
3. SDS-PAGE elettroforesi e dosaggio di attività dell’enzima;
4. Western blot;
5. Determinazione dei parametri cinetici enzimatici, KM e Vmax
Prerequisiti
Nozioni di base di biochimica
Modalità didattica
Esperienze pratiche di laboratorio condotte in laboratori appositamente attrezzati.
Materiale didattico
Il materiale didattico dell’insegnamento sarà prevalentemente costituito da protocolli e dispense preparate dai docenti che verranno consegnati agli studenti all’inizio dell’attività didattica e resi disponibili sulla piattaforma e-learning.

Modalità di verifica del profitto e valutazione
La modalità di verifica prevede una prova scritta da svolgersi in aula informatica mirata alla valutazione delle competenze acquisite per ciascun modulo. Verrà valutata la capacità di elaborare quanto appreso tramite le esperienze pratiche nell’ambito di tutte le discipline e la capacità di esporre collegamenti interdisciplinari.
La prova sarà suddivisa in 6 sezioni, una per ogni modulo, con domande aperte, esercizi e domande a scelta multipla. Per poter superare l’esame è necessario che lo studente abbia una valutazione maggiore o uguale a 18 in tutte le discipline. Nel caso lo studente non raggiunga la sufficienza anche in una sola disciplina, la prova dovrà essere nuovamente sostenuta per intero. La prova avrà una durata complessiva di 2 ore.

Aims
This module aims at providing students with skills for the basic biochemical techniques: protein purification, qualitative and quantitative characterization, enzymatic activity assays.

At the end of the course, the student should be able to apply the knowledge acquired in the field of biochemistry, with particular emphasis on enzyme purification and functional characterization.

At the end of the course the student will be able to apply the knowledge acquired in the field of biochemistry

The student should be able to process what he has learned, how to propose the methods used and when they should be applied.

At the end of the course the student will be able to express himself appropriately in the description of the topics addressed with language properties and exposure safety.

At the end of the course the student will be able to reproduce the techniques adopted in similar contexts.
Contents
Protein purification and characterization: quantitative protein determination, estimation of purity and molecular weight, enzymatic activity assay.
Detailed program
This module includes 5 practical classes:
1.Preparation of the crude extract;
2.Enzyme purification by affinity chromatography and protein quantification by colorimetric assay; 3. SDS-PAGE electrophoresis and enzymatic activity evaluation;
4.Western blot;
5. Experimental determination of enzymatic parameters KMand VmaxAC
Prerequisites
General notions of biochemistry are needed.
Teaching form
Lab experimental activities in equipped labs.
Textbook and teaching resource
Slides and experimental protocols will be provided to students at the beginning of the teaching activity, and uploaded on the moodle teaching Platform.

Assessment method
Written test focussed on all teaching modules: the exam will be aimed at the evaluation of acquired competences in all disciplines involved. The ability to elaborate and integrate the experimental work with the theoretical basis of the experiments, and the development of interdisciplinary links will be evaluated.
The assessment will be organised in six sections, with open questions and multiple choice tests. In order to pass the exam it is necessary that the student has an evaluation greater than or equal to 18 in all the disciplines. In the event that the student does not achieve sufficiency even in one discipline, the test must be re-supported in full. The duration of the assessment will be 2 hours.
