Obiettivi
Gli obiettivi formativi generali del corso sono finalizzati a fornire le competenze di citologia, istologia ed embriologia.
Contenuti sintetici
Il corso ha come finalità principale la conoscenza dell'ultrastruttura della cellula, dell'organizzazione dei tessuti e dello sviluppo embrionale. Nel corso si affronteranno anche alcune correlazioni cliniche.
Programma esteso
CITOLOGIA
Introduzione. Tecniche di studio in istologia e colorazioni istologiche.
Membrana cellulare, struttura, ultrastruttura e composizione chimica, funzioni e proprietà.
Specializzazioni della superficie cellulare (cilia, stereocilia, flagello, microvilli), struttura, ultrastruttura.
Ultrastruttura e funzioni delle giunzioni cellulari (giunzioni occludenti, aderenti e comunicanti). Molecole di adesione.
Meccanismi di trasporto attraverso la membrana cellulare. Endocitosi ed esocitosi.
Organizzazione del nucleo interfasico: involucro nucleare e pori nucleari; lamina nucleare; nucleoplasma; cromatina; nucleolo.
Struttura e ultrastruttura dei ribosomi. Cenni sulla sintesi proteica e sulle principali modificazioni post-traduzionali.
Struttura, ultrastrutttura e funzioni del reticolo endoplasmico (rugoso e liscio).
Struttura, ultrastrutttura e funzioni dell'apparato di Golgi. Cenni sul destino delle proteine prodotte.
Lisosomi: biogenesi, struttura, ultrastruttura e funzioni. Eterofagocitosi ed autofagocitosi.
Perossisomi: ruttura, ultrastruttura e funzioni.
Mitocondri: struttura, ultrastruttura e funzioni.
Citoscheletro: microfilameneti, microtubuli e filamenti intermedi, struttura, ultrastruttura e funzioni.
ISTOLOGIA
Tessuti: caratteristiche generali e classificazione.
Struttura, ultrastruttura e funzioni di:
Epiteli di rivestimento;
Epiteli ghiandolari. esempi di ghiandole esocrine e modalità di secrezione (merocrina, apocrina, olocrina ed eccrina); esempi di ghiandole endocrine.
Tessuto connettivo propriamente detto. Sostanza extracellulare dei tessuti connettivi (sostanza fondamentale e fibre). Biosintesi del collagene. Cellule del tessuto connettivo lasso, denso e reticolare.
Tessuto adiposo (uniloculare e multiloculare);
Cartilagine (ialina, elastica e fibrosa);
Tessuto osseo (non lamellare e lamellare, compatto e trabecolare).
Tessuto muscolare (striato scheletrico, striato cardiaco e liscio). Ultrastruttura del sarcomero e meccanismo della contrazione.
Tessuto nervoso (neuroni e cellule gliali). Mielina e mielinizzazione. Fibre nervose;
Sangue ed emopoiesi
EMBRIOLOGIA
Introduzione. Gametogenesi (spermatogenesi e spermatozoo, ovogenesi e oociti).
Capacitazione.
Fecondazione. Reazione corticale. Zigote.
Prima settimana di sviluppo embrionale: segmentazione, morula, cavitazione, blastocisti (embrioblasto e trofoblasto).
Impianto.
Primi stadi di sviluppo dell’embrione umano: formazione dell’epiblasto e dell’ipoblasto, disco embrionale bilaminare.
III-IV settimana di sviluppo embrionale: linea primitiva, transizione epitelio-mesenchimale (gastrulazione umana), formazione del mesoderma, disco embrionale trilaminare, notocorda ed assi corporei, neurulazione (tubo neurale e cellule della cresta neurale).
I tre foglietti embrionali (ectoderma, endoderma e mesoderma) e loro derivati.
Somiti e loro derivati.
Ripiegamento cefalo-caudale e trasversale.
Sviluppo della cavità celomatica.
Prerequisiti
Conoscenze scientifiche a livello di scuola secondaria superiore.
Modalità didattica
Il corso prevede 20 ore totali di didattica frontale (2 ore ciascuna), svolte in modalità erogativa in presenza.
Le lezioni saranno svolte in lingua inglese.

The general educational objectives of the course are aimed at providing cytology, histology and embryology skills.
Contents
The main aims of the course are the knowledge of the ultrastructure of the cell, the organization of tissues and of the embryonic development. Some clinical correlations will also be addressed during the course.

CYTOLOGY
Introduction. Techniques employed in histology and histological staining.
Cell membrane (structure, ultrastructure and chemical composition).
Specializations of the cell surface (cilia, stereocilia, flagellum, microvilli).
Ultrastructure and functions of cell junctions (tight, adherent and gap junctions). Adhesion molecules.
Transport mechanisms across the cell membrane. Endocytosis and exocytosis.
Organization of the interphase nucleus: nuclear envelope and nuclear pores; nuclear lamina; nucleoplasm; chromatin; nucleolus.
Structure and ultrastructure of ribosomes. Notes on protein synthesis and the main post-translational modifications.
Structure, ultrastructure and functions of the endoplasmic reticulum (rough and smooth).
Structure, ultrastructure and functions of the Golgi complex.
Fate of the proteins produced.
Lysosomes: biogenesis, structure, ultrastructure and functions. Heterophagocytosis and autophagocytosis.
Peroxisomes, structure, ultrastructure and functions.
Mitochondria: structure, ultrastructure and functions.
Cytoskeleton: microfilamenets, microtubules and intermediate filaments.
HISTOLOGY
General characteristics and classification of huma tissues.
Structure, ultrastructure and functions of:
- Lining epithelia;
- Glandular epithelia. examples of exocrine glands and ways of secretion (merocrine, apocrine, holocrine and eccrine); examples of endocrine glands
- Connective tissue proper. Extracellular substance of connective tissues (ground substance and fibres). Collagen biosynthesis. Cells of loose, dense and reticular connective tissue.
- Adipose tissue (unilocular and multilocular);
- Cartilage (hyaline, elastic and fibrous);
- Bone tissue (non-lamellar and lamellar, compact and trabecular).
- Muscle tissue (striated skeletal, striated cardiac and smooth). Ultrastructure of the sarcomere and mechanism of contraction.
- Nervous tissue (neurons and glial cells). Myelin and myelination. Nerve fibres;
- Blood and hematopoiesis
EMBRYOLOGY
Introduction. Gametogenesis (spermatogenesis and sperms, oogenesis and oocytes).
Capacitation. Fertilization. Cortical reaction. Zygote.
First week of embryonic development: segmentation, morula, cavitation, blastocyst (embryoblast and trophoblast).
Implantation.
First stages of development of the human embryo: formation of the epiblast and hypoblast, bilaminar embryonic disc.
III-IV week of embryonic development: primitive streak, epithelial-mesenchymal transition (human gastrulation), mesoderm formation, trilaminar embryonic disc, notochord and body axes, neurulation (neural tube and neural crest cells).
The three embryonic layers (ectoderm, endoderm and mesoderm) and their derivatives.
Somites and their derivatives.
Embryo folding.
Development of the coelomic cavity..
Prerequisites
Scientific knowledge at high school level
Teaching form
10 Frontal lessons (2 hours each), in attendance.
